Manager Lee Attenborough brought Ben Rhodes back into the starting lineup while club captain Jack Broadhead took a place on the bench, with Nathan Whitehead slotting into the back four. Toby Gregory also returned to the squad and would make his first appearance since early November.
Belper dominated the first ten minutes and to be fair were comfortably the better team in the first half but when chances came they were too often missed.
Having said that, Glossop had a good spell of their own and used it to take the lead. A warning signal had been sent in the 10th minute when Belper gave the ball away but Richard Brodie's shot was over the bar.
They did however take the lead in the 16th minute when from a corner Keaton Mulvey was on hand at the back post to volley home.
Belper restored parity only two minutes later when after a cross came in, the unfortunate Phil Perry stuck his leg out and saw the ball bounce into his own net.
A quick break down the right on 25 minutes saw Ben Rhodes put in a pinpoint cross onto the head of Luke Rawson but the ball clipped the crossbar and went over.
In the 34th minute the Nailers were forced into a change when Nathan Whitehead hobbled off and was replaced by Paulo Aguas.
Lewis Austin had an effort from just outside the box in the 36th minute which went just over.
The final action of the first half saw a long ball from the back reach Keaton August, his cross found Rawson but the shot was scuffed just wide.
The second half was slow to get going for both sides and the first action of note came in the 68th minute when a nice move for Glossop involving Dylan McMahon, Tom Lawless and Richard Brodie ended with the latter heading just wide of the post.
Belper took the lead in the 74th minute when Aguas's cross from the left reached Ben Rhodes. His half-volley was perfectly timed and placed into the bottom corner.
However, like Belper in the first half Glossop levelled almost immediately. When a cross was swung in, Abdul Djolo was first to the ball and his header squared things up at 2-2.
Four minutes from time, some superb passing play between Gregory, Aguas and Crofts brought a shooting opportunity for Crofts but Alex Brown produced a good save.
In the second minute of injury time, a cross from August on the right found Rawson's head but Brown pulled off another great stop as the ball looped down.
Glossop had the final chance to win the match in normal time when Belper conceded possession and substitute Kyle Worsley ran through but his shot was over the bar.
With that the match went to penalties, with Belper shooting first but missing two of their first four. Glossop had chances to win the shootout with their fifth and sixth penalties but Nailers goalkeeper Kieran Preston pulled off saves from Brodie, Worsley and finally McMahon to send Belper through, winning the shootout 4-3.
